The north London outfit found themselves a goal behind after just three minutes at the Britannia stadium as Steven Nzonzi headed City into an early lead.
However Clint Dempsey equalised for Andre Villas-Boas' side and Charlie Adam was sent off just five minutes after the half-time break which gave Spurs added belief they could claim victory.
Up stepped Adebayor as he hit his second goal in a week, giving Spurs a crucial three points in their quest to achieve Champions League qualification.
Adebayor was full of praise for his sides victory, as he told reporters: "It was an important game and we all knew we had to come here and win.
"If we kept passing the ball around we knew they would tire, that's what happened and we got our reward in the end."
Tottenham now hold a two point advantage over their north London rivals Arsenal. With a game in hand, the Gunners host Wigan Athletic at the Emirates tomorrow evening in a crunch game for both sides.
